A US team claim to have produced the first evidence for endogenous formation of tobacco-specific nitrosamines in rats. After 4 days of administration (twice-daily by gavage) of nicotine and sodium nitrite (at 10 and 12.4 mg/kg body weight, respectively), N′-nitrosonornicotine,N ′-nitrosoanabasine and N′-nitrosoanatabine were detectable in the urine. The investigators considered that the endogenous formation of these three compounds is likely in smokers and “may contribute significantly to their body burden of nitrosamines”
