Peptide nucleic acid probe-based fluorescence melting curve analysis for rapid screening of common JAK2, MPL, and CALR mutations

Detection of JAK2, MPL, and CALR mutations is essential in BCR-ABL1-negative MPN. Peptide nucleic acid-based fluorescence melting curve analysis is developed. The limit of detection (LOD) was approximately 10% for each mutation. Interference reactions using mixtures of different mutations were not observed. The rapid and simple PNA-based FMCA is a sensitive and specific technique.
