eIF2alpha kinases control the response to proteotoxic stress. Distinct eIF2alpha kinases are important in different cell types. eIF2a phosphorylation promotes and inhibits apoptosis. ROS and Ca2+ mediate proteotoxicity-induced apoptosis. Constitutive eIF2alpha phosphorylation may serve as a biomarker in cancers.
