The LRRK2 kinase domain can use GTP for both autophosphorylation and trans peptide phosphorylation activity. The pathological mutation G2019S increased GTP-dependent kinase activity. LRRK2 utilization of GTP as a phospho-donor changes the preferences of LRRK2 autophosphorylation.
