Shared featured pharmacophore modeling was explored on six brain receptors underlying the pathophysiology of migraine. Virtual screening and molecular docking indicated nystatin to be the lead active against maximum number of receptors. Brain targeted nystatin encapsulated chitosan nanoparticles were formulated and characterized by in-vitro and in-vivo studies. The in-silico results were validated through in-vivo studies performed on brain targeted nystatin nanoparticles. Nystatin was affirmed to be a multi-targeted novel lead for the therapeutic management of migraine.
