Selecting optimal columns for clarithromycin impurity analysis according to the quantitative relationship of hydrophobic subtraction model

A column selection method was proposed to identify optimal columns for specific separations of real samples. Quantitative relationships for critical pairs in clarithromycin impurity analysis were built based on HSM. Separation factors of the critical pairs of solutes were predicted accurately according to the above quantitative relationships. Ten desired columns from a database of more than 500 columns were identified and their column parameters were similar. The drawbacks of conventional distance-based column selection procedure are overcome.
