Sphingosine 1-phosphate (S1P) activates S1PR2 as well S1PR1 in osteoblasts.

Activation of S1P/S1PR2 promotes the signaling such as RhoA and the kinase (ROCK).

Activation of S1P/S1PR2/RhoA/ROCK signaling induces phosphorylation of Smad1/5/8.

The phosphorylation induces the expression of Runx2 and alkaline phosphatase.

S1P/S1PR2 signaling is important for osteogenesis in vivo as well as in vitro.
