The induction of T cell nonresponsiveness by blocking the CD40- and CD86-costimulatory molecules

Introduction: For complete T cell activation, costimulatory signals are required. Amongst multiple costimulatory ligand pair interactions, the interaction between CD80/CD86 on APC and CD28 on T cells is considered as the most important one, because CD28 generates an intracellular signal distinct from the TCR signaling pathway, and which prevents anergy induction. Expression of CD86 and more clearly of CD80 is regulated through CD40 triggering. In this study, blocking the CD40-CD40L interaction was therefore combined with CD80/CD86-blocking agents, and the consequences for allogeneic stimulation and anergy induction were analysed in MLR experiments between freshly isolated peripheral blood mononuclear cells (PBMC).
