Modulation of phagocytic function of bovine mononuclear phagocytes byHaemophilus somnus

The interactions between bovine mononuclear cells andHaemophilus somnusare known to be complex. To study this interaction, a flow cytometric assay was developed to assess the effect ofH. somnuson phagocytosis of killed opsonizedStaphylococcus aureusby bovine alveolar macrophages and blood monocytes. Using thisin vitrosystem, it was found that log phaseH. somnussignificantly inhibited the phagocytosis of killed opsonizedS. aureusby bovine alveolar macrophages obtained both from healthy calves and from cattle experimentally infected withH. somnus. However, killed log-phaseH. somnus,in vitropassaged and stationary phaseH. somnushad no effect on the phagocytic activity of these cells. In contrast to bovine alveolar macrophages, blood monocytes showed a significant increase in their phagocytic activity followingin vitroexposure to either log or stationary phaseH. somnus. Using a lypophilic, non-toxic fluorophore PKH2 to label liveH. somnus, it was possible to simultaneously measure the uptake of bothS. aureusandH. somnus. Stationary and log phaseH. somnuswere taken up by macrophages equally well, even though phagocytosis ofS. aureuswas inhibited by only log phaseH. somnus. These results demonstrate the ability ofH. somnusto modulate bovine mononuclear phagocytic function which might contribute towards the pathogenesis of bovine hemophilosis.
