O-glycosylation of rice-cell-derived recombinant human GM-CSF was investigated.

Eight O-glycans comprising four Ara with zero to seven Gal were determined.

O-glycans are heterogeneously attached to Ser 5, Ser 7, Ser 9, or Thr 10 residues.

Glycans on rrhGM-CSF are essential to the stability of its α-helix structure.

This is the first report on O-glycosylation of human GM-CSF derived from plant cells.
