Rational method for selecting influential amino acids on catalytic efficiency and thermostability of Suc2 invertase (SInv) was explained.

Five novel mutants of SInv were developed via site-directed mutagenesis and subjected to physicochemical examinations.

A novel mutant (P152 V) of SInv with enhanced catalytic efficiency was introduced.

A novel mutant (S305V-N463 V) of SInv with increased thermostability was introduced.

The influential impacts of Pro152 and Lys153 in accommodating sucrose in SInv active site were demonstrated.
