Defines and characterizes the catalytic domain of human APOBEC3B (A3B) enzyme.

Identifies a structural role for a conserved methionine at the junction of amino terminal and carboxy terminal domains.

Determines kinetic constants for the catalytic domain and shows that the amino terminal domain is inactive.

Determines the selectivity of A3B for cytosine compared to 5-methylcytosine.

These results imply that A3B is unlikely to play a major role in mutating or demethylating 5-methylcytosines in human genome.
