HepG2 cells stably expressing CYP3A4 were generated by modified piggyBac system.

The enzyme activity of CYP3A4 sustained stably and highly over 40 passages.

The cell lines were more sensitive to the toxic effects of bioactive compounds.

The cell lines could be suitable for predicting CYP3A4-mediated hepatotoxicity.
