In sheep with no signs of clinical paratuberculosis Faecal real time PCR applied for the detection of MAP DNA before parturition may result to significant underestimation of positivity compared to the periods after parturition and before the mating season. Real time PCR performed on one faecal sample collected from each animal after parturition, detected 71.4% of all PCR-positive animals. ELISA-testing resulted to an underestimation of positivity of 57%, referenced to faecal real time PCR. The selection of the period before parturition for the detection of shedders by faecal real time PCR is probably not ideal.
