An LC–MS/MS based method for 25(OH)D2 and 25(OH)D3 quantification was validated.

A complete stability profile of standard solutions and QC samples was obtained.

No differences were found between matching serum, heparin, and EDTA plasma samples.

25(OH)D concentrations measured were close to the DEQAS LC–MS/MS Method Means.

The LC–MS/MS method showed limited correlation to DiaSorin Liaison 25(OH)D assay.
