We observed non-toxic concentrations of multi-targeted kinase inhibitor sorafenib clearly impaired virus production.
Sorafenib markedly inhibited the activity of EV71 IRES, first step of the EV71 genomic RNA replication and translation.
Sorafenib prevented both EV71-induced CPE and the activation of ERK and p38 related to up-regulation COX-2/PGE2 expression.
We conclude that sorafenib inhibits EV71 replication by regulating viral IRES-dependent translation.
