Cord blood (CB) mononuclear cells were expanded with artificial antigen-presenting cells. Natural killer (NK)-cell expansion, phenotype, and cytotoxicity against B-cell non-Hodgkin lymphoma were evaluated. Expanded NK-cell cytotoxicity was increased compared with controls in vitro. Expanded NK cells were associated with decreased tumor burden and increased survival in vivo. The potential future use of expanded NK cells as adoptive cellular immunotherapy after umbilical CB transplantation was explored.
