Microgonotropens consist of a minor groove binding moietyfunctionalized with alkylamine side chains which are protonatedand positively charged at neutral pH and capable of formingelectrostatic bonds with the negatively charged phosphodiesterbackbone of DNA. We have investigated the interactions betweenmicrogonotropens and double-stranded DNA by a combination offluorescence, ultraviolet, and nuclear magnetic resonance spectroscopy. Our work reveals why microgonotropens are such potentinhibitors of the binding of transcription factors to their DNAbinding sites.
