The presence of trace contamination of soil and groundwater with explosives is an ongoing concern, for whichimproved methods are required to facilitate their detectionand quantification. This is true both for the monitoring ofremediation and for site characterization. Immunosensorshave been found effective for solution-phase detection ofenvironmental contaminants. Our work utilized the Luminex100 (flow cytometer) to detect TNT in a multiplexeddisplacement immunoassay format. The Luminex100 canperform a multiplexed assay by discriminating betweenup to 100 different bead sets. We used this capability toevaluate four different TNT monoclonal antibodies, tworecombinant TNT antibodies, and a control antibodysimultaneously for the rapid detection of TNT and othernitroaromatics. TNT could be detected at 0.1 ppb andquantified over the range of 1.0 ppb to 10 ppm. Inaddition, the assay was shown to be effective in variousmatrixes such as lake water, seawater, and acetoneextracts of soil. Seawater required dilution with two partsbuffer to avoid loss of microspheres, while the acetoneextracts were diluted 100-fold or more to minimizesolvent affects.
