Confocal micro-Raman spectroscopy requires a minimumof sample handling and no reagents and allows fastidentification of microorganisms. Since it reflects theoverall molecular composition of the cells, it providesmuch more information than classical, microbial analyses.However, since the molecular makeup of a cell dependson culture conditions, it can be argued that this will affectthe reproducibility and discrimination ability of Ramanspectroscopy. We used Bacillus cereus, Bacillus pumilus, and Bacillus licheniformis, which are known to beclearly distinct from each other and each displayingimportant phenotypic heterogeneity, in a wide variety ofculture conditions to analyze this. It is illustrated that theinfluence of culture conditions on the identification accuracy and taxonomic resolution of Raman spectroscopyis important though the effect on the final identificationis limited within the set of stains studied. Furthermore,some conditions even allow for better discrimination thanothers. From a practical point of view, it is especiallyimportant that differences in culturing time (and culturingtemperature) can be accommodated.
