The fern Pteris vittata accumulates unusually high levelsof arsenic. Using X-ray absorption spectroscopy (XAS) andXAS imaging, we reveal the distribution of arsenicspecies in vivo. Arsenate is transported through thevascular tissue from the roots to the fronds (leaves), whereit is reduced to arsenite and stored at high concentrations.Arsenic-thiolate species surrounding veins may beintermediates in this reduction. In gametophytes, arseniteis compartmentalized within the cell vacuole. Arsenic isexcluded from cell walls, rhizoids, and reproductive areas.This study provides important insights into arsenichyperaccumulation, which may prove useful for phytoremediating arsenic-contaminated sites, and demonstratesthe strengths of XAS imaging for distinguishing highlylocalized species.
