The rapid chemical analysis of individual cells is ananalytical capability that will profoundly impact manyfields including bioaerosol detection for biodefense andcellular diagnostics for clinical medicine. This articledescribes a mass spectrometry-based analytical techniquefor the real-time and reagentless characterization ofindividual airborne cells without sample preparation. Wecharacterize the mass spectral signature of individualBacillus spores and demonstrate the ability to distinguishtwo Bacillus spore species, B. thuringiensis and B.atrophaeus, from one another very accurately and fromthe other biological and nonbiological background materials tested with no false positives at a sensitivity of 92%.This example demonstrates that the chemical differencesbetween these two Bacillus spore species are consistentlyand easily detected within single cells in seconds.
