Antimutagenic Activity of (+)--Eudesmol and Paeonol from Dioscorea japonica

A methanol extract from Dioscorea japonica showed asuppressive effect on umu gene expression ofthe SOS response in Salmonella typhimuriumTA1535/pSK1002 against the mutagen 2-(2-furyl)-3-(5-nitro-2-furyl)acrylamide (furylfuramide). The methanolextract from D. japonica was re-extracted with dichloromethane, n-butanol, and water,respectively. The suppressive compoundsin the dichloromethane extract fraction were isolated bySiO2 column chromatography and identifiedas (+)--eudesmol and 2-hydroxy-4-methoxyacetophenone (paeonol) byEI-MS and 1H and 13C NMRspectroscopy. (+)--Eudesmol suppressed the SOS-inducingactivity of furylfuramide in the umutest. Gene expression was suppressed 80% at <0.18 mol/mL, andthe ID50 value was 0.09 mol/mL. Paeonol also suppressed the SOS-inducing activity offurylfuramide in the umu test. Geneexpression was suppressed 60% at <1.2 mol/mL, and theID50 value was 0.99 mol/mL. (+)--Eudesmol and paeonol were assayed with the mutagen3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole(Trp-P-1), which requires liver metabolizing enzymes, and showedsuppressive effects on the SOSinduction of the mutagen. (+)--Eudesmol suppressed geneexpression 48% at <0.18 mol/mL.Paeonol suppressed gene expression 76% at <1.2 mol/mL, andthe ID50 was 0.41 mol/mL. Theantimutagenic activities of these compounds against furylfuramide andTrp-P-1 were tested by anAmes test using S. typhimurium TA100, whichindicated that these compounds showed antimutagenic activity.Keywords: Dioscorea japonica; Dioscoreaceae;Chinese yam; (+)--eudesmol; paeonol;antimutagenicactivity; umu test; Ames test
