Biocatalysis is the activity to synthesize chemicals using biological entities, in particular enzymes. Natural enzymes often needto be optimized before being used in industrial processes, anddirected evolution is an approach that is well suited for thecreation of these improved industrial biocatalysts. Directedevolution involves creating a library of mutant genes and thensorting them based on the associated phenotypes. Main librarycreation and sorting protocols are compared here, with anemphasis on semirational approaches to library creation andfast screening or pool selection techniques. Representativereports of improved catalytic activity, enzyme stability, andspecificity are discussed. Finally, an insight is given on recenttechnological developments to make enzymes more suitable forbiocatalysis. These new technologies could favor the switch ofnumerous processes from chemistry to biocatalysis soon.
