Influence of Coupling Method on the Luminescence Properties, Coupling Efficiency, and Binding Affinity of Antibodies Labeled with Europium(III) Chelates

A series of chelating 4-(phenylethynyl)pyridines having various1,3,5-triazin-2-ylamino groups at thepara position of the phenyl ring was synthesized. Theireuropium chelates were coupled to antibodiesand the properties of antibody conjugates analyzed by fluorometry andin time-resolved fluorometricimmunoassay. The substituents in the triazine ring were observedto have various effects on thechelate luminescence, the labeling properties of the chelates, and theimmunoreactivity of labeledantibodies. The series of substituted triazinyl derivatives servesas a model of bioreactive groupsthat can be applied when certain properties are searched for, such asimproved chelate solubilities,minimized internal quenching, different effects on the ligand tripletstate, and stipulated couplingreactivities.
