We demonstrate the use of laser-induced fluorescenceconfocal spectroscopy to measure analyte-stimulated enhanced green fluorescent protein (egfp) synthesis bygenetically modified Escherichia coli bioreporter cells.Induction is measured in cell lysates and, since thespectroscopic focal volume is approximately the size ofone bioreporter cell, also in individual live bacteria. Thisis, to our knowledge, the first ever proof-of-concept workutilizing instrumentation with single-molecule detectioncapability to monitor bioreporter response. Although weuse arsenic inducible bioreporters here, the method isextensible to gfp/egfp bioreporters that are responsive toother substances.
