Characterization of the Stress Response of a Bioluminescent Biological Sensor in Batch and Continuous Cultures

The effects of temperature, growth stage, and inducer (ethanol)concentration on thekinetics and magnitude of the stress response were investigated byusing anEscherichia coli strain with the grpE heat shockpromoter fused to the Vibrio fischerilux genes. When stressed, the cells responded bychanging the level of specific lightemission, which was measured both on- and off-line. Thesemeasurements were usedto characterize and optimize the sensitivity of the construct bydetermining theconditions at which the culture exhibited maximum specificbioluminescence andminimum response time to ethanol induction in batch cultivation.The results of thebatch study were then applied to continuous cultivation, and the effectof dilutionrate was determined. These results are of considerable interest inthe developmentof an on-line biological sensor system for the detection and toxicityassessment ofchemical pollutants.
