Identification of small-molecule ligands for a protein ofinterest can facilitate the analysis of the protein's functionsin biological systems. Small-molecule microarrays haveallowed for rapid detection of such ligand-protein interactions in a high-throughput manner, although a label ona protein is needed to observe these interactions. Bycombining SPR imaging technology with our recentlydeveloped photo-cross-linked small-molecule array platform, we developed a novel platform that allows in situobservation of interactions between photo-cross-linkedsmall molecules on gold surfaces and nonlabeled proteinsin solution. Interactions of estrogenic and androgenicsubstances with estrogen receptor were observed usingthis platform.
