Direct observation of single native DNA molecules in amicrochannel was monitored without fluorescence-dyelabeling. At a PDMS/glass microchip, the image of individual -DNA molecules appear sharp and distinct inNomarski differential interference contrast microscopy.Intercalator dyes affected the physical properties anddynamic behavior of individual DNA molecules. From themigration velocities in the microchannel it is evident thatnative DNA molecules migrated faster than DNA molecules labeled with the intercalator YOYO-1. This isbecause YOYO-1 increases the molecular weight and sizeof -DNA and decreases the charge. The electric fieldstrength and pH also affected the dynamics of single DNAmolecules. We also observed that YOYO-labeled DNA wasmore stretched out compared to native DNA.
