Stable red astaxanthin-producing transformants wereobtained after genetic transformation of two Phaffia rhodozymamutants. A yellow mutant, accumulating &bgr;-carotene, and an albino mutant,accumulating phytoene, from P. rhodozyma weretransformed using a genomic library of wild-typestrain UCD 67-385 in the pBluescript vector.Hybridization assays, using the pBluescript DNA as aradioactive probe, indicate integration of vectorsequences into the genome of the transformants.Transformants DNA was digested with restrictionendonucleases, ligated with T4 DNA ligase and thenused to transform E. coli. Ampicillinresistant plasmids, containing 0.1, 0.2, and 2.5 kbDNA inserts of P. rhodozyma, were rescued fromthe yeast red transformants. The molecular analysisindicate that transformation has occurred by anintegration event of donor DNA into the genome of thehost strains.
