Development of Near-Infrared Reflectance Spectroscopy (NIRS) Calibration Model for Estimation of Oil Content in Brassica juncea and Brassica napus

Vegetable oil is an important part of human diet since ancient days. Rapeseed-mustard is the third most important source of vegetable oil in the world. In the present study, the main aim was to develop calibration equation for oil content using near-infrared reflectance (NIRS) in Brassica juncea and Brassica napus. The reference values of oil content were generated on nuclear magnetic resonance (NMR). NIRS is a multitrait technique, i.e., a large number of traits can be measured simultaneously. Reflectance spectra of intact seeds of same samples were recorded (400–2500 nm) on NIR systems Model 6500 spectrophotometer (Foss-NIRSystem). The NIRS manipulation for scanning, mathematical processing, and statistical analysis was performed using WinISI software. Scoring was done to remove the redundant data. Spectra were treated as outliers if their global H was >3.0. Cross validation was done using modified partial least square method. The internal validation was done to test the goodness of fit of the developed equation. The oil content in the calibration sets varied from 33.8–44.6 % in case of B. juncea, 32.0–42.2 % in case of B. napus whereas RSQ values for oil content was 0.83 in B. juncea and 0.41 in B. napus indicated usefulness of calibration equation for the oil content. The predicted model validated in case of B. juncea with r2 value 0.85. The results indicated that NIRS could be used satisfactorily for rapid determination of oil content in B. juncea.
