Effects of dietary supplementation of Lactobacillus Plantarum on survival, hematological profile and histopathology of selected organs of Clarias gariepinus fingerlings

This study evaluates the effects of dietary supplementation of Lactobacillus plantarum (LP) on survival of hematological profile and histopathology of selected organs of Clarias gariepinus fingerlings. Ten Clarias gariepinus fingerlings were allocated to five treatments, with three replicated subgroups each, fed and reared for 12 weeks. LP isolated from corn slurry was cultured using standard procedures. Five iso-nitrogenous diets were formulated at 35 % crude protein (T0, T1, T2, T3, and T4) with LP at inoculation rate of 0 × 107 (T0), 0.5 × 107 (T1), 1.0 × 107 (T2), 1.5 × 107 (T3), and 2.0 × 107 (T4) cfu/g feed, respectively. Fish selected per treatment were challenged with Pseudomonas aeruginosa and monitored for 21 days. Packed Cell Volume (PCV), Red Blood Cell (RBC), Hemoglobin (Hb), and White Blood Cell (WBC) were determined as well as histopathology of the jejunum, liver, and kidney using standard procedures. Results showed that the PCV, RBC, and Hb values were high in T3, while the lowest values were observed in T1. The highest Heterophil-Lymphocytes ratio was recorded in T3 and T4. The treatment groups showed extremely low mortality rate compared to the control when challenged. Also, villus hypertrophy and increased thickness of laminar propria due to development of gut associated lymphoid tissue was pronounced in T4 & T3, mildly prominent in T2 & T1 and absent in T1. With increasing DS, hepatocellular tubular epithelial degeneration, tubular necrosis and glomerular mesangial cell loss were observed more in T0, T1, T2, and mildly in T3 & T4. These results indicated that the use of LP in diets as probiotics confer protection against pathogens in C. gariepinus.
