The effects of acute 2 mg sublingual 17β-estradiol on exercise induced coronary ischemia in older women with coronary artery disease

The effects of acute administration of 2 mg sublingual 17β-estradiol (E2) on exercise induced coronary ischemia in postmenopausal women were studied. The patients (n=12) had >1 mm horizontal or downsloping ST-segment depression on a previous exercise test and stable angina pectoris with angiographically documented coronary stenosis (>50%). After sublingual administration of 2 mg of E2, or 2,5 mg nitroglycerine (NTG) or placebo in a randomised, cross over, double blind design the patients underwent a new symptom limited bicycle exercise test. The plasma levels of E2 increased from 62±26 to 4243±1485 pmol/L after 30 minutes. Estrone and estronesulphate levels were measured. E2 did not induce any improvement in total exercise time (Ext), or time onset of ST-segment depression (STd), when compared to NTG and P. Results shown in table are expressed as mean values ±SD* p<0.05 vs P. Heart rate × SBP = heart rate × systolic blood pressure × 103.
