Six new cardenolides, periforosides D-E (rong class="boldFont">1rong>-rong class="boldFont">2rong>), periforgenin C (rong class="boldFont">3rong>) and periforosides F-H (rong class="boldFont">4rong>-rong class="boldFont">6rong>), as well as 10 previously identified cardenolides (rong class="boldFont">7rong>-rong class="boldFont">16rong>) were isolated from the ethanol extract of the stems of Periploca forrestii. The structures of the new compounds were determined using extensive spectroscopic analyses including HRESI-MS, 1D and 2D NMR data. Evaluation of the cytotoxic activity of all the isolated compounds in five different human cancer cell lines indicated that compounds rong class="boldFont">2rong>-rong class="boldFont">6rong>, rong class="boldFont">8rong>, rong class="boldFont">9rong> and rong class="boldFont">12rong>-rong class="boldFont">16rong> have potent activity.
