The susceptibility of different Populuseuramericana (Neva, PE68-022 x P. nigra, 71-060 x P. nigra)and P. deltoides (PE68-022 x P. deltoides) clones towild-type Agrobacterium tumefaciens strains(A281 and 82.139) was evaluated in an inoculationexperiment, and differences in the frequency of tumorformation (0-48) were found. Co-cultivationexperiments demonstrated high transformation abilityof oncogenic binary A. tumefaciens strains ascompared to disarmed strains. Using oncogenic binarystrains, transgenic calluses were obtained from alltested clones. The presence of acetosyringone did notinfluence the transformation frequency of the disarmedstrains. Co-inoculation experiments were performedusing leaf discs and a bacterial suspension containingboth wild-type and disarmed strains. No positiveeffects on transformation efficiency were noticed inthese conditions either. The transformation of tumorsand kanamycin resistant calluses was confirmed by DNAanalysis.
