摘要
为了研究我国新型新城疫病毒的分子流行病学特性,对2016年从主动监测中分离到的1株新出现的鸭源新城疫病毒(duck/China/NX2209/2016)进行了基因组测序和遗传进化分析。序列分析结果显示该分离株全长15 198nt,F基因ORF全长为1 662bp,共编码553个氨基酸,裂解位点处的氨基酸为:~(112) ERQERL~(117),符合低致病性新城疫病毒特征。同源性分析表明该新型毒株同北美分离株具有较高的同源性。F基因高变区进化分析结果显示,该病毒属于ClassⅠ,但不属于基因型1~10,为新型毒株。完整F基因进化树结果显示,该病毒属于基因1c亚型,与北美分离株高度同源。
        In order to understand the molecular epidemiology of Newcastle disease virus in China,we sequenced and analyzed one new emerging ClassⅠ NDV(duck/China/NX2209/2016)isolated from duck in 2016 during the active surveillance.Sequence analysis revealed that genome of the ClassⅠ NDV was 15 198 nt in length.The length of complete open reading frame of the F gene was 1 662 bp and encoding 553 amino acids.The putative amino acid at the cleavage site of F protein was~(112)ERQERL~(117),showed typical molecular characteristics of low pathogenic Newcastle disease virus.Homology analysis indicated the new isolate had higher homology with the NDVs isolated from North America.Genetic analysis based on the main functional region of F gene revealed that the virus was new emerging in China and didn't belong to any identified known genotype(genotype 1-10).The phylogenetic tree based on the complete F gene showed that the virus belonged to the 1cgenotype and showed highly homologous with the viruses isolated from wild birds in North American.
