摘要
本研究以膨珊瑚茎段为外植体,采用正交试验设计,进行芽诱导离体培养研究;并应用徒手切片技术观察同化枝的形态解剖结构,分析其对生境的适应性。研究表明:膨珊瑚茎段最佳灭菌处理为75%酒精30 s+0.1%升汞11 min,最佳外植体为半木质化茎段(上段)或顶端;芽诱导的最佳培养基为1/2 MS+6-BA1.2 mg/L+NAA 0.1 mg/L;芽诱导效果最佳为半木质化茎段(上段);芽继代增殖最佳培养基为3/4 MS+ZT1.0 mg/L+NAA 0.02 mg/L+GA3 0.3 mg/L;膨珊瑚叶片退化,茎枝具有同化功能,皮层及髓部发达,含有较大的薄壁细胞,供其贮藏营养物质与水份。
        In this study, the stem segments of Euphorbia oncoclada were used as explants, and orthogonal experimental design was applied to study the in vitro culture of bud induction. The morphological and anatomical structure of the assimilating branch was observed by using the technique of freehand section, which was used to analyze the adaptability of Euphorbia oncoclada to habitats. Results showed that the best sterilization treatment for the stem segments of Euphorbia oncoclada was 75% alcohol for 30 s and 0.1% mercuric chloride for 11 min, and the best explant was semi-lignification stem upper segment or tip stem. Moreover, the best medium for bud induction was 1/2 MS+6-BA 1.2 mg/L +NAA 0.1 mg/L, the best effect of bud induction was semi-lignification stem upper segment, and the best medium for bud subculture was 3/4 MS+ZT 1.0 mg/L+NAA 0.02 mg/L+GA30.3 mg/L. The leaves of Euphorbia oncoclada have been degenerated, but its stem and branch have assimilation function, and its cortex and pith are well developed, which contain large parenchyma cells for storage of nourishment and water.
