摘要
本实验选用"Fondant"、"Pink Surprise"、"China Pink"和"White Pearl"4个品种鳞片分化出脱毒试管苗的鳞片叶和小鳞茎作为外植体,探究植物激素对风信子试管苗分化的影响。实验结果表明,试管苗鳞片叶直接诱导分化不定芽的能力强于小鳞茎,前者最适宜的细胞分裂素浓度分别为:6-BA 0.5 mg/L (Fondant)、TDZ 0.1 mg/L (China Pink)、6-BA 1.0 mg/L (White Pearl)和6-BA 0.5 mg/L (Pink Surprise),后者最适宜的细胞分裂素浓度分别为:ZT 0.5 mg/L (Fondant)、TDZ 0.1 mg/L (China Pink)、TDZ 0.4 mg/L (White Pearl)和ZT1.5 mg/L (Pink Surprise);试管苗鳞片叶最适宜的植物激素配比为6-BA 0.5 mg/L+IBA 0.5 mg/L (Fondant)、6-BA 0.5 mg/L+IBA 0.5 mg/L (Pink Surprise)、6-BA 1.0 mg/L+IBA 0.5 mg/L (White Pearl);小鳞茎最适宜的植物激素浓度比例为:ZT 0.5 mg/L+NAA 0.5 mg/L (Fondant)、ZT 1.5 mg/L+NAA 0.5 mg/L (Pink Surprise)、TDZ0.4 mg/L+IBA 1.0 mg/L (White Pearl);"Fondant"和"White Pearl"的最适生根浓度为NAA 1.5 mg/L和NAA1.0 mg/L。本研究为风信子组织培养体系的优化提供了科学依据。
        In this experiment, the squama leaves and bulblets of virus-free plantlets derived from 4 varieties of squama, "Fondant", "Pink Surprise", "China Pink" and "White Pearl", were used as explants to explore the effects of plant hormones on the differentiation of hyacinth plantlets in vitro. The experimental results showed that the ability of directly inducing differentiation adventitious buds from squama leaves of test-tube plantlets was stronger than that of bulblets. The most suitable cytokinin concentrations for the former were 6-BA 0.5 mg/L(Fondant),TDZ 0.1 mg/L(China Pink), 6-BA 1.0 mg/L(White Pearl), and 6-BA 0.5 mg/L(Pink Surprise). For the latter, the most appropriate cytokinin concentrations were ZT 0.5 mg/L(Fondant), TDZ 0.1 mg/L(China Pink), TDZ 0.4 mg/L(White Pearl), and ZT 1.5 mg/L(Pink Surprise). The most suitable plant hormones proportions in squama leaves of test-tube plantlets were 6-BA 0.5 mg/L+IBA 0.5 mg/L(Fondant), 6-BA 0.5 mg/L+IBA 0.5 mg/L(Pink Surprise),and 6-BA 1.0 mg/L+IBA 0.5 mg/L(White Pearl). And for the small bulbs, the most appropriate plant hormone concentration are ZT 0.5 mg/L+NAA 0.5 mg/L(Fondant), ZT 1.5 mg/L+NAA 0.5 mg/L(Pink Surprise), and TDZ 0.4 mg/L+IBA 1.0 mg/L(White Pearl). In addition, the research showed that NAA 1.5 mg/L and NAA 1.0 mg/L were the optimum auxin concentrations for "Fondant" and "White Pearl". This study might provide a scientific basis for the optimization of hyacinth tissue culture system.
