摘要
为了更好地评价不同产地苍耳子标准汤剂的出膏率、绿原酸含量及含量转移率,对苍耳子标准汤剂的质量进行了研究。采用日本Shimadzu LC-20A型高效液相色谱仪,Agilent Zorbax Eclipse C_(18)色谱柱(2.1 mm×250 mm,5.0μm),以乙腈-磷酸为流动相,柱温为28℃,流速为1 mL/min,检测波长为327 nm,建立了苍耳子标准汤剂中绿原酸含量的测定方法。结果表明:15批苍耳子标准汤剂出膏率的均值为4.30%,均值±3SD为3.28%~5.32%;绿原酸含量均值为3.44 mg/g,均值±3SD为0~7.16 mg/g;绿原酸含量转移率均值为36.54%,均值±3SD为0.66~72.42%;不同产地苍耳子的出膏率较为稳定,但绿原酸含量及含量转移率差异较大。经方法学考察,苍耳子标准汤剂中绿原酸含量测定方法稳定,具有较高的重复性、稳定性及可信度,可用于苍耳子标准汤剂的质量评价,为苍耳子配方颗粒后续研究及大规模生产提供数据支持。
        In order to better evaluate the extraction rate, chlorogenic acid content and content transfer rate of Xanthii Fructus standard decoction in different producing areas, the quality of standard decoction of Xanthii Fructus is studied. The Japanese Shimadzu LC-20 A high performance liquid chromatograph and Agilent Zorbax Eclipse C_(18) column(2.1 mm × 250 mm, 5.0 μm) are applied in the method. The mobile phase is acetonitrile-phosphoric acid solution, the column temperature is 28 ℃, the flow rate is 1 mL/min, and the detection wavelength is 327 nm. A method is established for determining the content of chlorogenic acid in Xanthii Fructus standard decoction. The results show that the average yield of 15 batches of Xanthii Fructus standard decoction is 4.30%, and the mean value ±3 SD ranges from 3.28% to 5.32%.The average chlorogenic acid content is 3.44 mg/g, and the mean ±3 SD ranges from 0 to 7.16 mg/g. The average chlorogenic acid content transfer rate is 36.54%, and the mean value ±3 SD ranges from 0.66 to 72.42%. The results show that the extraction rate of Xanthii Fructus in different habitats is stable, and the chlorogenic acid content and content transfer rate are different. In view of methodology, the method for the determination of chlorogenic acid in Xanthii Fructus standard decoction is stable with high repeatability, stability and credibility, and can be used for quality evaluation of Xanthii Fructus standard decoction. This method provides data support for the follow-up research and mass production of Xanthii Fructus formula granules.
