猪CISH基因过表达对IL-6和TNF-α基因mRNA的表达调控研究

摘要
为了分析猪细胞因子诱导的含SH2结构域蛋白(CISH)基因对白细胞介素6(IL-6)和α肿瘤坏死因子(TNF-α)等基因表达的调控,深入研究CISH基因的生物学功能,试验采用猪CISH基因过表达质粒pcDNA-CISH瞬时转染PK-15细胞,利用Real-time PCR技术分析了CISH过表达对相关基因表达量的影响。结果表明:在PK-15细胞中,CISH基因过表达并没有引起TLR4、MyD88、NF-κB、IL-1β、IL-6、TNF-α等基因相对表达量的显著改变;细菌脂多糖(LPS)刺激PK-15细胞后,IL-6、TNF-α和TLR4等基因的相对表达量极显著上调(P<0.01);LPS刺激CISH基因过表达的PK-15细胞后,IL-6基因的相对表达量显著上调(P<0.05)。说明在LPS的刺激下,CISH过表达可以上调IL-6基因的相对表达量。
