摘要
目的探讨雪菊和杭白菊的水提物对凝血功能和血小板聚集的影响。方法 80只昆明小鼠随机分为8组:正常对照组(生理盐水)、溶媒组(0.5%CMC-Na)、阿司匹林高、低剂量组(90、30 mg/kg)、雪菊水提物高、低剂量组(20、10 g/kg)、杭白菊水提物高、低剂量组(20、10 g/kg),每组10只。40只SD大鼠随机分为5组:正常对照组(生理盐水)、溶媒组(0.5%CMC-Na)、阿司匹林组(15 mg/kg)、雪菊水提物组(10 g/kg)和杭白菊水提物组(10 g/kg),每组8只。采用煎煮法得到雪菊和杭白菊的水提物,比较各组凝血时间、出血时间、凝血四项、体内、外血小板聚集率的差异。结果雪菊水提物高剂量组(20 g/kg)、杭白菊水提物、高低剂量组(20、10 g/kg)均可以显著延长小鼠的凝血时间和出血时间(P<0.01)。雪菊水提物可以延长凝血酶时间(TT)(P<0.05),杭白菊水提物可以延长活化部分凝血活酶时间(APTT)(P<0.05)。在体外实验中,雪菊和杭白菊的水提物均可以降低由二磷酸腺苷(ADP)、凝血酶、花生四烯酸(AA)诱导的血小板聚集的聚集率(P均<0.01)。在体内实验中,雪菊水提物组可以显著降低由ADP诱导的血小板聚集率,杭白菊水提物组可以显著降低由AA诱导的血小板聚集率(P<0.01)。结论雪菊和杭白菊的水提物具有抗凝血和体内体外的抗血小板聚集作用。
        Objective To explore the effects of the water extracts of Coreopsis tinctoria and Chrysanthemum morifolium on coagulation function and platelet aggregation. Methods 80 Kunming mice were randomly divided into eight groups: normal control group(saline), solvent group(0.5% CMC-Na), aspirin high and low dose group(90, 30 mg/kg), Coreopsis tinctoria water extract high and low dose group(20, 10 g/kg), Chrysanthemum morifolium water extract high and low dose group(20, 10 g/kg), 10 mice in each group. 40 SD rats were randomly divided into five groups: normal control group(saline), solvent group(0.5% CMC-Na), aspirin group(15 mg/kg), Coreopsis tinctoria water extract group(10 g/kg) and Chrysanthemum morifolium water extract group(10 g/kg), 8 rats in each group. The water extracts of Coreopsis tinctoria and Chrysanthemum morifolium were obtained by decoction. The differences of coagulation time, bleeding time, four coagulation items, and platelet aggregation rate in vivo and in vitro were compared. Results The high dose group(20 g/kg) of Coreopsis tinctoria water extract, the high dose group(20 g/kg) and the low dose group(10 g/kg) of Chrysanthemum morifolium water extract could significantly prolong the clotting time and bleeding time of mice(P<0.01). The Coreopsis tinctoria water extract could prolong the time of thrombin(TT)(P<0.05), and the Chrysanthemum morifolium water extract could prolong the time of activated partial thromboplastin(APTT)(P<0.05). In vitro, the water extracts of Coreopsis tinctoria and Chrysanthemum morifolium could reduce the platelet aggregation rate induced by adenosine diphosphate(ADP), thrombin and arachidonic acid(AA)(P<0.01). In vivo, the platelet aggregation rate induced by ADP was significantly decreased in the water extract group of Coreopsis tinctoria, and that induced by AA was significantly decreased in the water extract group of Chrysanthemum morifolium(P<0.01). Conclusion The water extracts of Coreopsis tinctoria and Chrysanthemum morifolium have the effects of anticoagulation and anti-platelet aggregation.
