摘要
目的研究中药提取物对羊布鲁菌的抗菌活性,为临床羊布鲁菌感染患者选用中药治疗提供实验依据。方法使用VITEK 2 Compact全自动微生物鉴定仪及16S rDNA基因序列分析对临床分离的羊布鲁菌进行菌种鉴定;采用微量肉汤稀释法测定紫草、虎杖及钩藤等28种中药提取物对羊布鲁菌的最低抑菌浓度(minimal inhibitory concentration,MIC)。结果VITEK 2 Compact全自动微生物鉴定仪及16S rDNA基因序列分析的细菌鉴定结果均为羊布鲁菌(Brucella melitensis)。28种中药提取物中,紫草、虎杖、钩藤对羊布鲁菌的MIC为15. 6 mg/m L;女贞子、山豆根、连翘、黄精、白芍的MIC为31. 25 mg/m L;黄药子、玄参、厚朴、益母草、瓜菱的MIC为62. 5 mg/m L;龙胆草、菌味、百部、木香的MIC为125 mg/m L;苦瓜、天冬、杜仲的MIC为250 mg/m L;猪牙皂、拳参、三七、木蝴蝶、姜黄、白芨的MIC为500 mg/m L;半夏、黄柏的MIC> 1000 mg/m L。结论 28种中药提取物中,除半夏和黄柏外对羊布鲁菌都具有一定程度的抗菌作用,以紫草、虎杖和钩藤对羊布鲁菌的抗菌效果最好,女贞子、山豆根、连翘、黄精、白芍次之。因此,临床治疗羊布鲁菌感染患者,可选用抗菌效果较好的中药辅助治疗,减少抗菌药物长时间使用,避免药物的耐药性及毒副作用的产生。
        Objective This study was aimed at observing the antibacterial activities of Chinese herbal extracts on brucella melitensis,providing experimental evidence for patients infected with brucella melitensis to select traditional Chinese medicine in clinical treatment. Methods The brucella melitensis separated from clinic was identified again by Vitek 2 Compact automatic microorganism identification instrument and 16 S rDNA gene sequence analysis,separately. Micro broth dilution method was used to determine the minimal inhibitory concentration of twenty eight Chinese medicine extracts like Lithospermum erythrorhizon,Polygonum cuspidatum and Uncaria rhynchophylla against Brucella melitensisin in vitro. Results Among the twenty eight Chinese herbal extracts,the MIC of Lithospermum erythrorhizon,Polygonum cuspidatum and Uncaria rhynchophylla were 15. 6 mg/ml,respectively;The MIC of Fructus ligustri lucidi,radix Sophorae tonkinensis,Forsythia suspensa,Polygonatum sibiricum and Cynanchum otophyllum were 31. 25 mg/ml; The MIC of Dioscoreaceae,radix scrophulariae,Magnolia officinalis rehd,leonurus Artemisiaand melon rhombus were 62. 5 mg/ml,respectively; The MIC of Elephantopusscaber,bacterial taste,stemona japonica and common vladimiria rootwere 125 mg/ml,respectively; The MIC of momordica charantia L.,radix asparagiand eucommia ulmoides were 250 mg/ml,separately; The MIC of Fructus gleditsiae,Polygonum bistorta L.,panax notoginseng,oroxylum indicum,Curcuma longa L. and Buddleja alternifolia maxim were 500 mg/ml,respectively; The MIC of Pinellia ternate and Cortex phellodendri were higher than 1 000 mg/ml. Conclusion Twenty eight kinds of Chinese herbal extracts all have an antibacterial effect on Brucella melitensis except for Pinellia ternateand cortex phellodendri. Lithospermum erythrorhizon,Polygonum cuspidatum and Uncaria rhynchophylla have the best antibacterial effect,followed by Fructus ligustri lucidi,radix Sophorae tonkinensis,Forsythia suspensa,Polygonatum sibiricum and Cynanchum otophyllum. Therefore,patients infected with Brucella melitensis may choose traditional Chinese medicine of better antibacterial effects,so as to shorten antibacterial medication time,avoid drug resistance,toxic and side effects.
