头穴丛刺法对阿尔茨海默病大鼠学习记忆能力及海马CA1区炎症介质的影响

摘要
目的:观察头穴丛刺法对阿尔茨海默病(AD)大鼠学习记忆能力及海马CA1区炎症介质的影响;方法:40只符合标准的SPF级Wistar大鼠随机分为空白组、假手术组、模型组和头穴丛刺组,每组10只。模型组和头穴丛刺组通过双侧海马区注射Aβ1-42溶液复制AD模型,假手术组注射等量无菌双氧水,空白组不做处置。头穴丛刺组于造模后2 d起给予针刺干预,空白组、假手术组和模型组仅背侧位捆绑固定,1次/d,连续治疗14 d。治疗结束后通过Morris水迷宫实验检测AD大鼠学习记忆能力,ELISA法检测海马CA1区中Ach、ChAT和AchE的含量和活性,免疫组化和RT-PCR法分别检测海马CA1区中IL-6、IL-10蛋白和mRNA的表达;结果:与空白组相比,模型组逃避潜伏期较空白组明显增加,跨越平台次数和有效停留时间较空白组明显降低,海马组织中Ach的含量、ChAT的活性、IL-10蛋白及mRNA的表达较空白组明显降低,AchE的活性和IL-6蛋白及mRNA的表达较空白组明显增加,差异均具有统计学意义(P<0.05)。与模型组比较,头穴丛刺能够显著降低大鼠逃避潜伏期时间,增加跨越平台次数和有效停留时间,增加海马组织中Ach的含量、ChAT的活性、IL-10蛋白及mRNA的表达,降低AchE的活性和IL-6蛋白及mRNA的表达,差异均具有统计学意义(P<0.05)。结论:头穴丛刺疗法能够通过调控AD大鼠海马CA1区组织中炎症因子的表达和促进Ach的含量,从而改善AD大鼠的学习记忆能力。
        Objective: To observe the effects of cluster needling of scalp point therapy on learning and memory ability and inflammatory factors in hippocampal CA1 region of rats with Alzheimer's disease(AD). Methods: 40 SPF Wistar rats were randomly divided into control group, sham-operated group, model group and acupuncture group with 10 rats in each group. The AD model was replicated by injecting β-amyloid protein 1-42 solution into bilateral hippocampus in model group and acupuncture group. The sham-operated group was injected with the same amount of sterile hydrogen peroxide, while the control group was not disposed. The acupuncture group was treated with cluster needling of scalp point therapy after model establishment. The control group, sham operation group and model group were bundled and fixed in the dorsal position once a day for 14 consecutive days. After treatment, the learning and memory abilities of AD rats were tested by Morris water maze test, the contents and activities of Ach, ChAT and AchE in hippocampal CA1 region were detected by ELISA, and the expressions of IL-6, IL-10 protein and mRNA in hippocampal CA1 region were detected by immunohistochemistry and RT-PCR, respectively. Results: Compared with the control group, the escape latency of the model group was significantly increased(P<0.05), the number of crossing platforms and effective residence time were significantly decreased(P<0.05), the content of Ach, the activity of ChAT, the expression of IL-10 protein and mRNA in hippocampus were significantly decreased(P<0.05), and the activity of AchE and the expression of IL-6 protein and mRNA were significantly increased(P<0.05). Compared with the model group, cluster needling of scalp point therapy could significantly reduce the escape latency time(P<0.05), increase the number of crossing platforms and effective residence time(P<0.05), increase the content of Ach, the activity of ChAT, the expression of IL-10 protein and mRNA in hippocampus(P<0.05), and decrease the activity of AchE and the expression of IL-6 protein and mRNA(P<0.05). Conclusion: Scalp acupoint cluster needling therapy can improve the learning and memory ability of AD rats by regulating the expression of inflammatory mediators and promoting the content of Ach in hippocampal CA1 region of AD rats.
