摘要
目的:观察小儿久咳合剂对感染后咳嗽(肺热阴虚证)大鼠咳嗽敏感性及血清中IL-5、TNF-α、IgE含量的影响,探讨该药降低咳嗽敏感性,减轻气道炎性反应的作用机制。方法:从50只SD大鼠中随机选取10只作为空白组,其余40只采用烟熏+内毒素滴鼻+甲状腺素灌胃+辣椒素雾化诱咳法建立感染后咳嗽(肺热阴虚证)大鼠模型,造模成功后随机分为模型组、孟鲁司特钠组、小儿久咳合剂小剂量组及小儿久咳合剂大剂量组,每组10只。各组分别予以相应药物进行干预,空白组及模型组则均给予同体积的0.9%氯化钠溶液灌胃。多功能诱咳引喘仪检测各组大鼠咳嗽敏感性,ELISA法检测治疗前后各组大鼠血清中IL-5、TNF-α、IgE水平。结果:与模型组比较,小儿久咳合剂大、小剂量组大鼠咳嗽频次明显减少,血清中IL-5、TNF-α、IgE水平也明显降低(P<0.05,P<0.01)。结论:小儿久咳合剂能降低模型大鼠的咳嗽敏感性,减少模型大鼠血清中IL-5、TNF-α和IgE等炎性介质的释放,从而减轻气道炎性反应。
Objective: To observe the effects of Pediatric Chronic Cough Mixture(PCCM) on cough sensitivity and the content of IL-5, TNF-α and IgE in post infectious cough(PIC) rats with syndrome of yin deficiency due to lung heat and explore the mechanism. Methods: The fifty rats were enrolled and randomly divided into control group(n=10) and model group(n=40). The PIC(syndrome of yin deficiency due to lung heat) model rats was established by cigarette-smoking exposure, intranasal administration with small dose of LPS, thyroid hormone and capsaicin atomization method. Then, the model rats were enrolled and randomly divided into model group(n=10), montelukast sodium group(n=10), large PCCM group with low doses(n=10) and high doses(n=10). 0.9%Saline, montelukast sodium suspension and PPCM was given to the rats in corresponding group as the therapeutic drug. When the treatment was finished, the cough sensitivity and the contents of IL-5, TNF-α and IgE in serum of rats were determined. Results: Compared with the model group, the cough frequency and the content of IL-5, TNF-α and IgE in serum of rats in large PCCM group with low doses and high doses were declined(P<0.05, P<0.01). Conclusion: PPCM can reduce cough sensitivity of the model rats. PPCM might treat PIC(syndrome of yin deficiency due to lung heat) by reducing the release of IL-5, TNF-α, IgE and other inflammatory mediators in serum.
