雨生红球藻破壁孢子粉对小鼠酒精性损伤肾脏的保护作用研究

摘要
为了研究雨生红球藻破壁孢子粉对酒精性损伤小鼠肾脏的保护作用,将小鼠随机分为正常对照组、模型组和雨生红球藻粉低、中、高剂量组(100 mg/kg、200 mg/kg、400 mg/kg)。其中,剂量组动物先灌胃受试物7 d,第8天开始,给予剂量组和模型组50%酒精连续灌胃7 d,建立酒精性损伤模型。最后一次灌胃16 h后处死小鼠,取血清和相关脏器,测定小鼠血清尿素氮(blood urea nitrogen, BUN)和肌酐(creatinine, CREA)浓度,同时测定肾脏中超氧化物歧化酶(superoxide dismutase, SOD)活性以及谷胱甘肽(glutathione, GSH)、丙二醛(malondialdehyde, MDA)、肿瘤坏死因子-α(tumor necrosis factor-α, TNF-α)和白细胞介素-1β(interleukin-1β, IL-β)含量。结果显示:与模型组相比,雨生红球藻破壁孢子粉各剂量组肾脏指数降低, BUN和CREA含量显著降低(P<0.01),SOD活性和GSH含量显著升高(P<0.01), MDA显著降低(P<0.01),炎性因子TNF-α和IL-β也显著降低(P<0.05)。上述实验结果表明雨生红球藻破壁孢子粉对小鼠酒精性肾脏损伤具有明显的保护作用。
        To explore the kidney protective effect of Haematococcus pluvialis on alcoholic injury, mice were randomly divided into the control group, model group and groups treated with different doses(100 mg/kg,200 mg/kg and 400 mg/kg) of the H. pluvialis powder. The alcoholic liver injury model was induced by intragastric administration with 50% ethanol for 7 days. About 16 h after the last administration, the animals were killed, and the sera and kidneys were obtained for determining the related markers, including the blood urea nitrogen(BUN) and creatinine(CREA), and superoxide dismutase(SOD), glutathione(GSH), malondialdehyde(MDA), tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) in kidney. Compared with the model group, mice in treatment groups had a reduced kidney index, significantly decreased levels of BUN and CREA(P<0.01), significantly increased SOD activity and contents of GSH(P<0.01), and significantly decreased contents of MDA(P<0.01), and also levels of inflammatory factors(TNF-α and IL-1β)(P<0.05). These results suggested that H. pluvialis had a significant kidney protective effect on alcohol-induced injury in mice.
