平肝脉通片对阴虚阳亢高血压大鼠一氧化氮、内皮素干预作用的研究

摘要
目的:探讨平肝脉通片对阴虚阳亢高血压大鼠一氧化氮(NO)、内皮素(ET)干预作用的研究。方法:采用二肾一夹方法并灌服附子汤制备阴虚阳亢高血压大鼠模,选择造模成功的75只大鼠,随机分为空白对照组、卡托普利对照组、平肝脉通片低剂量组、平肝脉通片中剂量组、平肝脉通片高剂量组,每组15只,分别给予2 m L/100g生理盐水、10 mg/m L卡托普利溶液及10、20、40 mg/m L的平肝脉通片溶液灌胃,连续4周,对比观察各组治疗前后尾动脉血压、血清NO、内皮素-1(ET-1)、血管紧张素Ⅱ(AngⅡ)、血浆醛固酮(ALDO)以及心功能指标左室射血分数(EF%)、短轴缩短率(FS%)变化。结果:与治疗前相比,治疗后空白对照组尾动脉血压、ET-1、AngⅡ、ALDO显著升高(P<0.05),NO显著降低(P<0.05),EF%、FS%无明显变化,而其他4组尾动脉血压、ET-1、AngⅡ、ALDO显著降低,NO、EF%、FS%显著升高,差异均有统计学意义(P<0.05),其中平肝脉通片高剂量组、中剂量组治疗后尾动脉血、ALDO显著低于托普利对照组,NO、EF%、FS%显著高于托普利对照组,治疗后平肝脉通片高剂量AngⅡ显著低于托普利对照组,差异均有统计学意义(P<0.05)。结论:平肝脉通片能有效降低阴虚阳亢高血压大鼠血压,逆转左心室肥厚,应与其能调节血清NO、ET-1、AngⅡ、ALDO有关。
        Objective: To explore the effect of nitric oxide,endothelin on the Yin deficiency type hand renal hypertensive rats intervened by Pinggan-Maitong slices. Methods: The deficiency type hand renal hypertensive rats has been made by adopting the method of two kidney one clip and irrigation clothing monkshood; 75 successfully built rats cases were selected,and they were randomly divided into blank control group,captopril group,hepatic vein on low dose group,hepatic vein on middle dose group,hepatic vein on high dose group,with 15 cases in each group. They were only given 2 m L /100 g physiological saline,10 mg / m L concentration of captopril solution,10 mg / m L,20 mg / m L,and 40 mg / m L concentration of hepatic vein through solution to fill the stomach respectively,for four weeks. Tail arterial blood pressure,serum NO,endothelin 1( ET-1),angiotensin Ⅱ( AngⅡ),plasma aldosterone( ALDO) and cardiac function index of left ventricular ejection fraction( EF) %,short axial shortening rate( FS) % between groups before and after treatment were observed. Results: The tail artery blood pressure,ET-1,AngⅡ in blank control group after treatment were significant rise,ALDO,NO significantly reduced,EF%,FS% had no obvious change,while the tail artery blood pressure,ET-1,Ang Ⅱ,ALDO in other four groups were significantly reduced,NO,EF%,FS% were significantly raised,differences were statistically significant( P < 0. 05); The tail artery blood pressure,ALDO in hepatic vein on middle or high dose group were significantly lower than the captopril control group,NO,EF%,FS% were significantly higher than the captopril control group,the AngⅡ in hepatic vein on high dose group was significantly lower than the captopril control group,the differences were statistically significant( P < 0.05). Conclusions: Pinggan-Maitong slice can reduce the blood pressure in yin deficiency type hand hypertension rats,it can reverse left ventricular hypertrophy,which may related to adjusting the serum NO,ET-1,Ang Ⅱ,ALDO.
