摘要
目的观察丹参多酚酸对脑缺血大鼠氧化应激的影响。方法将72只雄性SD大鼠随机分为Sham、MCAO和治疗组。每组分为1 d、7 d和14 d 3个子组,每子组8只。MCAO组和治疗组选用线栓法制备大鼠大脑中动脉闭塞模型,Sham组只分离血管不结扎。治疗组在线栓术3 h评分后即刻股静脉注射丹参多酚酸注射液(1.6 mg/kg),Sham和MCAO组同法给予等量生理盐水。每只大鼠在线栓术后3 h和处死前进行Longa神经功能评分,采用HE染色法观测神经元形态,采用化学比色法测定MDA水平和SOD活性。结果 Sham组SD大鼠神经缺损分值为0分;MCAO组和治疗组大鼠各子组给药前评分比较,差异无统计学意义(P>0.05);不同时间点分值比较,差异有统计学意义(P<0.05),线栓术后3 h神经缺损分值开始增加,术后1 d达峰,随后逐渐下降,差异有统计学意义(P<0.05),术后7、14 d治疗组分值均低于MCAO组,差异有统计学意义(P<0.05)。MCAO组、治疗组MDA水平高于Sham组,差异有统计学意义(P<0.05);术后7、14 d治疗组MDA水平低于MCAO组,差异有统计学意义(P<0.05)。MCAO组、治疗组SOD活性低于Sham组,差异有统计学意义(P<0.05);术后7、14 d治疗组SOD活性高于MCAO组,差异有统计学意义(P<0.05)。Sham组各时间点MDA水平和SOD活性比较,差异无统计学意义(P>0.05);MCAO组和治疗组MDA水平和SOD活性随时间延长逐渐下降,差异有统计学意义(P<0.05)。结论丹参多酚酸可能通过减少氧化应激损伤发挥脑保护作用。
        Objective To observe the oxidative stress injury intervention effect of salvia miltiorrhiza polyphenolic acid injection in brain of rats with cerebral infarction.Methods Seventy-two male sprague dawley(SD) rats were randomly divided into Sham group, MCAO group and treatment group. Each group was subdivided into 3 subgroups(1 d, 7 d and 14 d), 8 each. In the MCAO group and the treatment group, the model of middle cerebral artery occlusion in rats was prepared by suture thrombectomy. Sham group was just separated the blood vessels. Score was performed 3 h after operation and then injection of salvia miltiorrhiza polyphenolic acid(1.6 mg/kg) was administered intravenously in the treatment group. The Sham group and the MCAO group were treated with equal amount of normal saline. The Longa neural function score was performed 3 h after operation and before execution. The neuronal morphology was observed by HE staining. The MDA content and SOD activity were measured by chemical colorimetry.Results Rats in Sham group showed no neurological symptoms and score of 0. Scores before treatment were no statistical different among MCAO and treatment subgroups(P>0.05), and scores in different time were statistical significance(P<0.05). The neurological impairment function score 3 h after operation began to increase, peaked 1 day after the operation, and then gradually decreased, the difference was statistically significant(P<0.05). Scores of 7 d and 14 d treatment group were lower than MCAO group and the differences were statistically significant(P<0.05). The MDA levels in MCAO and treatment group were higher than Sham group, and the differences were statistical significance(P<0.05). It was lower in treatment group than MCAO group in 7 d and 14 d(P<0.05). The SOD activity in MCAO and treatment group was lower than Sham group, and the differences were statistical significance(P<0.05). It was higher in treatment group than MCAO group in 7 d and 14 d(P<0.05). The MDA levels and SOD activity at different times in Sham group were no statistically different(P>0.05). The MDA levels and SOD activity gradually decreased over time in MCAO and treatment groups, and the differences were statistically significant(P<0.05).Conclusion Salvianolate may play a protective role in brain by reducing oxidative stress injury.
