日钟阴阳方对LED灯照下兔视网膜多巴胺及其D2受体水平的影响

摘要
目的:探讨日钟阴阳方对LED灯照下兔视网膜多巴胺及其D2受体水平的影响。方法:1月龄健康无眼病新西兰幼兔24只随机分成LED灯组和LED灯中药组,每组12只。再将每组分成4周组和12周组2个亚组,每组6只。LED灯组幼兔饲养在LED灯光照控制箱内,自由饮水和进食。LED灯中药组幼兔饲养环境同LED灯组,同时采用"日钟阴阳方"灌胃。于实验前及实验后4、8、12周测量2组幼兔体质量。测定视网膜DA及D2受体水平。结果:实验前后4组幼兔体质量均无显著变化(P> 0. 05)。第4、12周时,LED灯中药组的视网膜DA水平显著高于LED灯组(P <0. 01),视网膜D2受体蛋白在上述视网膜细胞层内的表达及D2受体表达量吸光度值强于LED灯组(P <0. 05或P <0. 01),但4周组和12周组比较无显著差异(P> 0. 05)。结论:在LED灯照下,日钟阴阳方可显著提高视网膜中多巴胺及其受体的表达水平,具有保护视网膜组织结构的作用,值得临床推广。
        Objective:To explore the influence of Rizhong Yinfang Formula on levels of dopamine and its receptor D2 in retina of young rabbits under exposure of light-emitting diode( LED). Methods:A total of 24 oculopathy-free young New Zealand rabbits at the age of 1 month were selected and randomly divided into LED group and LED traditional Chinese medicine( TCM) group,12 cases in each group. Both groups were re-divided into two subgroups,4-week group and 12-week group,6 cases in each group. Young rabbits in LED group were fed with free intake of water and food under exposure of LED,on which basis LED TCM group added gastric perfusion of Rizhong Yinyang Formula. The body weight of rabbits in all groups were measured before experiment and 4 and 12 weeks after experiment. And then the levels of dopamine( DA) and its receptor D2 were detected. Results:There was insignificant difference among 4 groups in the body weight before and after experiment( P > 0. 05). LED TCM group was evidently higher in DA levels in retina than that of the LED group( P < 0. 01) and was notably stronger in the expression of D2 receptor protein in above retinal cell layers and the absorbance values of D2 receptor expression levels than those of the LED group( P < 0. 05 or P < 0. 01) after 4-week and 12-week experiment,but there was insignificant difference between 4-week and 12-week groups( P> 0. 05). Conclusion:Under exposure of LED,Rizhong Yinyang Formula can significantly increase the expression levels of DA and its receptors in retina and protect retinal tissue structures,and thus deserving to be popularized clinically.
