摘要
目的:建立高效液相色谱法(HPLC)测定大鼠各组织中没食子酸含量的方法,研究没食子酸在大鼠体内的组织分布特征。方法:大鼠按50 mg·kg~(-1)灌胃给予没食子酸后取各组织,采用沉淀蛋白法预处理生物组织样品,色谱柱:Fortis-C_(18)柱(250 mm×4.6 mm,5μm),流动相:甲醇-0.1%磷酸水=25∶75;检测波长273 nm;柱温30℃;进样体积20μl;流速1 ml·min-1,测定各组织中没食子酸的含量。结果:各组织中没食子酸的浓度在0.50~128.00μg·ml~(-1)范围内线性关系良好。灌胃0.5 h后各组织中均能检测到没食子酸,在各组织分布中,小肠中含量最高,肺中含量最低。结论:建立了大鼠组织中没食子酸的HPLC测定方法,该方法简便、灵敏度高、专属性强,结果准确;由此法测得没食子酸组织分布迅速且广泛,主要分布在小肠、肾、肝脏组织中。
        Objective: To establish a high performance liquid chromatography method for the determination of gallic acid in tissues of rats,and study the tissue distribution characteristics of gallic acid in rats. Methods: The tissues were obtained after the oral administration of 50 mg · kg~(-1) gallic acid,and prepared by protein precipitation. The analytical column was a Fortis-C_(18) column( 250 mm× 4. 6 mm,5 μm). The mobile phase was a mixture of methanol and 0. 1% phosphoric acid( 25 ∶ 75),the detection wavelength was set at 273 nm,the column temperature was 30℃,the injection volume was 20 μl,and the flow rate was 1 ml·min~(-1). Results: The calibration curves of gallic acid in the tissues were linear within the range of 0. 50-128. 00 μg·ml~(-1). Gallic acid could be detected out in all the tissues at 0. 5 h after the oral administration. The highest content of gallic acid was checked out in small intestine,and the lowest was detected out in the lung. Conclusion: An HPLC method was established to detect gallic acid in the tissues of rats. The method is simple,sensitive,specific and accurate. The distribution of gallic acid is quick and extensive mainly in small intestine,kidney and liver.
