摘要
目的:建立五汁饮中阿魏酸和多糖的含量测定方法并进行方法学考察,以便控制该制剂的质量。方法:五汁饮中阿魏酸定量分析采用HPLC法,使用Ultimate 3000色谱柱,固定相为碳十八键合硅胶,流动相为甲醇-0.5%乙酸水溶液(30∶70),流速1mL·min~(-1),运行时间30min,检测波长321nm,柱温30℃;五汁饮中多糖定量分析采用苯酚-硫酸比色法。结果:五汁饮中阿魏酸含量测定结果表明,阴性无干扰,仪器精密度RSD为0.96%,供试品溶液48h内稳定(RSD为1.31%),重复性试验RSD为1.14%(n=6),平均加样回收率(n=9)98.69%,RSD为1.0%。阿魏酸理论塔板数为4 000,分离度为1.65。五汁饮中多糖平均含量为63.75%(RSD=5.2%,n=6),五汁饮供试液在6h内稳定,平均加样回收率为103.4%,RSD=1.16%(n=6)。结论:HPLC测定五汁饮中阿魏酸含量专属性好,重复性好、准确、灵敏、简便、快速,可作为五汁饮中阿魏酸含量测定的检测方法;苯酚-硫酸比色法可用于五汁饮中多糖的含量测定,以控制该制剂质量。
Objective:To establish the determination method of ferulic acid and polysaccharide content in the Five Juices Decoction and to carry out the methodology,so as to control the quality of preparation.Methods:The quantitative analysis of ferulic acid in the Five Juices Decoction was carried out by using the Ultimate 3000chromatographic column.The stationary phase was carbon octade-the running time was 30min.The detection wavelength was 321nm;the column temperature was 30℃;the quantitative analysis of poly-content in the Five Juices Decoction showed that it was negative,without any interference.The instrument precision RSD was0.96%and the test solution was stable within 48h(RSD was 1.31%).Through repeatable test,RSD was 1.14%(n=6).The average sample recovery rate(n=9)was 98.69%and RSD was 1.0%.Besides,the number of theoretical plates was 4000,with the resolution of 1.65.In the Five Juices Decoction,the average content of polysaccharide was 63.75%(RSD=5.2%,n=6),with sta-tion of ferulic acid content in the Five Juices Decoction,HPLC has good specificity,good repeatability,accuracy,sensitivity,simplicity and rapidity.It can be used as a method for determination of ferulic acid content in the Five Juices Decoction.And phenol-sulfuric acid colorimetry can be used to determine the content of polysaccharide in the Five Juices Decoction so as to control the quality of this product.
